Three‐dimensional structured illumination microscopy (3D‐SIM) has opened up new possibilities to study nuclear architecture at the ultrastructural level down to the ∼100 nm range. We present first results and assess the potential using 3D‐SIM in combination with 3D fluorescence in situ hybridization (3D‐FISH) for the topographical analysis of defined nuclear targets. Our study also deals with the concern that artifacts produced by FISH may counteract the gain in resolution. We address the topography of DAPI‐stained DNA in nuclei before and after (...) 3D‐FISH, nuclear pores and the lamina, chromosome territories, chromatin domains, and individual gene loci. We also look at the replication patterns of chromocenters and the topographical relationship ofXist‐RNA within the inactive X‐territory. These examples demonstrate that an appropriately adapted 3D‐FISH/3D‐SIM approach preserves key characteristics of the nuclear ultrastructure and that the gain in information obtained by 3D‐SIM yields new insights into the functional nuclear organization. Upshot: The controversy between realism and constructivism often seems to be a matter of epistemology. However, empirical researchers are not primarily interested in solving philosophical questions but in practicing good research. It would be shortsighted to believe that there is a contradiction between epistemological and empirical questions.

Problem: First-order scientific research is often not aware of the hidden assumptions provided by an epistemological perspective based upon realism. Beyond philosophical considerations about the epistemological foundations, some practical normative implications deriving from them are crucial: in the field of communication and media studies, some scholars criticize media coverage, e.g., on climate change, as biased and distorted from reality. Method: From a constructivist perspective, the article presents a detailed meta-analysis of the course of argumentation provided by two empirical communication studies (...) that follow an objectivist approach. Result: With the help of a second-order research strategy, it is possible to uncover their ontological assumptions and criticize their normative implications. Implications: Social scientists should be careful with normative suggestions for the system under study unless they are applied within these systems themselves. (shrink)

Work of Eagle, Farah, Goldbring, Kirchberg, and Vignati shows that the only separable C*‐algebras that admit quantifier elimination in continuous logic are,,, and the continuous functions on the Cantor set. We show that, among finite dimensional C*‐algebras, quantifier elimination does hold if the language is expanded to include two new predicate symbols: One for minimal projections, and one for pairs of unitarily conjugate elements. Both of these predicates are definable, but not quantifier‐free definable, in the usual language of C*‐algebras. This paper seeks to encourage continued innovation in translating built environment and transportation-focused physical activity research into practice. Successful strategies, policies, and tools from across the U.S. and globally that demonstrate potential for wider-scale implementation are highlighted. The importance of building practice and translational research partnerships with groups and organizations outside traditional public health spheres, such as those who work in real estate and land-use development, is also discussed.

Das Thema greift aktuelle Debatten über Autorschaft, Urheberrecht, Originalität und Plagiat auf, die anzeigen, dass diese Konzepte neuerdings in Bewegung geraten sind. Die Beiträge des Bandes fragen aus unterschiedlichen Fachperspektiven, ob und inwiefern die Verfahren des Umgangs mit Autorschaft zwischen Mittelalter und Moderne am Ende stärker vergleichbar sind als gemeinhin angenommen. So werden die aus der Moderne gebildeten Kategorien von Kreativität hinterfragt und unterlaufen. (...) Zugleich zeigt sich, dass eben diejenigen Kreativitätsprozesse der Moderne, die den hohen normativen Ansprüchen einer Geniekultur nicht entsprechen, nicht einfach Defizienzerscheinungen sind, sondern Variationen vielfach geübter Techniken, zu deren Verständnis die mittelalterliche Produktivität dient. Schaffen und Nachahmen sind nicht in einen "künstlerischen" Bereich der Kultur sektorierbar, sondern sie erweisen sich als Grundstruktur menschlichen Handelns auch im wissenschaftlichen, ethischen oder religiösen Feld. What was classification as it first took modern form in the eighteenth century, how did it work, and how did it relate to earlier describing and ordering? We offer new answers to these questions by considering an example less well known than that of botany or zoology, namely medicine, and by reconstructing practice on paper. The first and best-known disease classification is the “nosology” of the Montpellier physician François Boissier de Sauvages de Lacroix. Its several editions, we show, were less (...) products than process: published tools for building a classification system. The disorder of a hitherto unstudied notebook that Boissier de Sauvages kept throughout this process provided a way of breaking with the topical order of earlier physicians’ humanistic commonplace books of disease observation while sustaining the paper practices those earlier physicians—and Sauvages himself as a student—had used to order disease. This suggests a different picture of historical change than that of a scholarly world of ordered words giving way to a scientific one of ordered things. Classification, in the case of Sauvages’ nosology, arose through an incomplete break with, and intensified practice of, a past way of ordering the described world. The humanist paper practice that had made observationes, differently applied, now made species. Classification into genera and species by similarity and difference, which Sauvages’ nosology shared with botany, was an algorithm of paper and ink practice—in its operation more machine-like than humanist textual practice yet in its effects more creative and re-creative of categories and questions of relationship. Thus a new empiricism of generalizations arose out of the older, Renaissance empiricism of particulars. (shrink)

The rapid expansion of research on Brain-Computer Interfaces is not only due to the promising solutions offered for persons with physical impairments. There is also a heightened need for understanding BCIs due to the challenges regarding ethics presented by new technology, especially in its impact on the relationship between man and machine. Here we endeavor to present a scoping review of current studies in the field to gain insight into the complexity of BCI use. By examining studies related to BCIs (...) that employ social research methods, we seek to demonstrate the multitude of approaches and concerns from various angles in considering the social and human impact of BCI technology. For this scoping review of research on BCIs’ social and ethical implications, we systematically analyzed six databases, encompassing the fields of medicine, psychology, and the social sciences, in order to identify empirical studies on BCIs. The search yielded 73 publications that employ quantitative, qualitative, or mixed methods. Of the 73 publications, 71 studies address the user perspective. Some studies extend to consideration of other BCI stakeholders such as medical technology experts, caregivers, or health care professionals. The majority of the studies employ quantitative methods. Recurring themes across the studies examined were general user opinion towards BCI, central technical or social issues reported, requests/demands made by users of the technology, the potential/future of BCIs, and ethical aspects of BCIs. Our findings indicate that while technical aspects of BCIs such as usability or feasibility are being studied extensively, comparatively little in-depth research has been done on the self-image and self-experience of the BCI user. In general there is also a lack of focus or examination of the caregiver’s perspective. (shrink)

This book critically assesses arguments for the existence of the God of classical theism, develops an innovative account of objects’ persistence, and defends new arguments against classical theism. The authors engage the following classical theistic proofs: Aquinas’s First Way, Aquinas’s De Ente argument, and Feser’s Aristotelian, Neo-Platonic, Augustinian, Thomistic, and Rationalist proofs. The authors also provide the first systematic treatment of the ‘existential inertia thesis’. By connecting the thesis to relativity theory and recent developments in the philosophy of physics, and (...) by developing a variety of novel existential-inertia-friendly explanations of persistence, they mount a formidable new case against classical theistic proofs. Finally, they defend new arguments against classical theism based on abstract objects and changing divine knowledge. The text appeals to students, researchers, and others interested in classical theistic proofs, the existence and nature of God, and the ultimate explanations of persistence, change, and contingency. It is widely recognized that motivation is an important determinant for a successful sports career. Specific patterns of motivational constructs have recently demonstrated promising associations with future success in team sports like football and ice hockey. The present study scrutinizes whether those patterns also exist in individual sports and whether they are able to predict future performance levels. A sample of 155 young individual athletes completed questionnaires assessing achievement goal orientations, achievement motives, and self-determination at t1. The person-oriented method linking (...) of clusters after removal of a residue was used to form clusters based on these motivational constructs in order to analyze the relations between these clusters and the performance level 2.5 years later. Similar to the studies in team sports, four motivational patterns were observed at t1. The highly intrinsically achievement-oriented athletes were much more likely to compete internationally [odds ratio = 2.12], compared to the failure-fearing athletes. Although team and individual sports differ in many respects, they nevertheless are characterized by similar and thus generalizable career-promoting motivational profiles: Regardless of the type of sport, the highly intrinsically achievement-oriented athletes consistently have the best potential for success. A brain-computer interface is a rapidly evolving neurotechnology connecting the human brain with a computer. In its classic form, brain activity is recorded and used to control external devices like protheses or wheelchairs. Thus, BCI users act with the power of their thoughts. While the initial development has focused on medical uses of BCIs, non-medical applications have recently been gaining more attention, for example in automobiles, airplanes, and the entertainment context. However, the attitudes of the general public towards BCIs have (...) hardly been explored. Among the general population in Germany aged 18–65 years, a representative online survey with 20 items was conducted in summer 2018 and analysed by descriptive statistics. The survey assessed: affinity for technology; previous knowledge and experience concerning BCIs; the attitude towards ethical, social and legal implications of BCI use and demographic information. Our results indicate that BCIs are a unique and puzzling way of human–machine interaction. The findings reveal a positive view and high level of trust in BCIs on the one hand but on the other hand a wide range of ethical and anthropological concerns. Agency and responsibility were clearly attributed to the BCI user. The participants’ opinions were divided regarding the impact BCIs have on humankind. In summary, a high level of ambivalence regarding BCIs was found. We suggest better information of the public and the promotion of public deliberation about BCIs in order to ensure responsible development and application of this potentially disruptive technology. This text looks at the potential benefits of concept and theory formation along dynamic, evolutionary and valuation for understanding economic processes.

In people with chronic low back pain, maladaptive structural and functional changes on a cortical level have been identified. On a functional level, somatosensory cortical excitability has been shown to be reduced in chronic pain conditions, resulting in cortical disinhibition. The occurrence of structural and/or functional maladaptive cortical changes in people with CLBP could play a role in maintaining the pain. There is currently no measurement protocol for cortical excitability that employs stimulation directly to the lower back. We developed a (...) protocol for the measurement of single pulse somatosensory evoked potential waveforms and paired-pulse behavior generated from sensory nerves of the lower back and quantified its test–retest reliability in a sample of 30 healthy individuals to gain insights into the normal variability of cortical responses, which could then be compared to results from people with CLBP. We investigated cortical excitability by measuring SEPs and PPB. PPB was defined as the ratio of the amplitude of the second cortical response divided by the first cortical response. A2s was determined by subtracting the response to single-pulse stimuli from the paired pulse stimuli response to account for linear superposition effects. The test–retest reliability of the protocol was very poor with no evidence of systematic bias but a high amount of random variability between sessions. There was no significant difference in the right side PPB for session 1 and session 2 ; mean session difference [ = −0.44 ; t = −1.17, p = 0.26]. The ICC3.1 for the outlier-removed right side PPB were 0.19 and 0.43 for left side PPB. This finding potentially has wider implications for PPB protocols. If these findings were replicated in other groups and other nerves, it would question the validity of this measure more generally. However, these findings are restricted to healthy people and sensory nerves of the lower back and may not be generalizable. (shrink)

This paper gives a survey of David Hilbert's (1862–1943) changing attitudes towards logic. The logical theory of the Göttingen mathematician is presented as intimately linked to his studies on the foundation of mathematics. Hilbert developed his logical theory in three stages: (1) in his early axiomatic programme until 1903 Hilbert proposed to use the traditional theory of logical inferences to prove the consistency of his set of axioms for arithmetic. (2) After the publication of the logical and set-theoretical paradoxes by (...) Gottlob Frege and Bertrand Russell it was due to Hilbert and his closest collaborator Ernst Zermelo that mathematical logic became one of the topics taught in courses for Göttingen mathematics students. The axiomatization of logic and set-theory became part of the axiomatic programme, and they tried to create their own consistent logical calculi as tools for proving consistency of axiomatic systems. (3) In his struggle with intuitionism, represented by L. E. J. Brouwer and his advocate Hermann Weyl, Hilbert, assisted by Paul Bemays, created the distinction between proper mathematics and meta-mathematics, the latter using only finite means. He considerably revised the logical calculus of thePrincipia Mathematica of Alfred North Whitehead and Bertrand Russell by introducing the ε-axiom which should serve for avoiding infinite operations in logic. Instead of arguing about elements and boundaries of civil society, recent discussions in social theory have focused on the concept of civil society itself as embedded in different currents of social and political thought. Following up on these discussions, this article reconstructs the concept of civil society by identifying a number of implicit oppositional terms and the respective semantic fields, which in different historical contexts have lent meaning to the concept. Three such oppositional terms and counter-meanings will be distinguished in (...) turn and traced back to different traditions of European social and political theory: (1) the barbarism of disorder; (2) the barbarism of order; and (3) the realm of toil and material necessity. It is argued that the multiple meanings and counter-meanings of civil society are connected by a deep structure of discourse. This deep structure of civil society thinking can be translated into a ‘semiotic square’ in the tradition of A.J. Greimas. In conclusion, it is suggested to further investigate current uses of civil society along these lines, in order to clarify normative goals and possible ways of mediating between opposing moral worlds. This chapter briefly summarises work by four key figures in the phenomenological philosophy of science: Edmund Husserl; Martin Heidegger; Patrick Heelan; and Joseph J. Kockelmans. In addition, some comparison is made with well-known figures in mainstream philosophy of science, and suggestions are given for further readings in the phenomenological philosophy of science.

What does it mean to practice socially responsible science on controversial issues? In a fresh turn focussing on the neuroscientists’ responsibility in producing knowledge about politically charged subjects, Chalfin et al. (Am J Bioethics 8(1):1–2, 2008) caution neuroscientists to be careful about how they present their findings lest their results be used to support unfounded biases, social stereotypes and prejudices. Weisberg et al. (J Cogn Neurosci 20(3):470–477, 2008) discuss the allure of neuroscience explanations and demonstrate how laypersons easily accept dubious (...) claims as long as (even non-relevant) neuroscientific descriptions are provided. Fine (2010) exposes the use of scientific evidence in propagating outdated gender myths by popular writers and discusses the infiltration of these prejudices into popular belief, folk culture and lifestyle. This paper discusses ways in which the ‘neuroscience of gender difference’ itself inadvertently contributes to normalising socially constructed theories about sex difference in cognitive performance. This unpremeditated effect has evident implications on the structuring of society because gender relations cut across social, political and economic boundaries. We present a theoretical reflection of factors that could interact with the scientists’ attempted objective evaluation of the subject, the methods and some principle problems, and we engage a science studies approach as our methodological tool. Our object of critique is drawn from the research on spatial abilities that articulate the dissention pertaining to sex differences in intellectual capacity. (shrink)

It is widely agreed that a new conception of history was developed in the early nineteenth century: the past came to be seen in a new light, as did the way of studying the past. This article discusses the nature of this collective revision, focusing on one of its first and most important manifestations: Ranke's 1824 Geschichten der romanischen und germanischen Völker. It argues that, in Ranke's case, the driving force of the revision was religious, and that, subsequently, an understanding (...) of the nature of Ranke's religious attitude is vital to any interpretation of his historical revision. Being aesthetic-experiential rather than conceptual or "positive," this religious element is reflected throughout Ranke's enterprise, in source criticism and in historical representation no less than in the conception of cause and effect in the historical process. These three levels or aspects of the historical enterprise correspond to the experience of the past, and are connected by the essence of the experience: visual perception. The highly individual character of the enterprise, its foundation in sentiments and experiences of little persuasive force that only with difficulty can be brought into language at all, explains the paradoxical nature of the Rankean heritage. On the one hand, Ranke had a great and lasting impact; on the other hand, his approach was never re-utilized as a whole, only in its constituent parts—which, when not in the relationship Ranke had envisioned, took on a new and different character. This also suggests the difference between Ranke's revision and a new paradigm: whereas the latter is an exemplary solution providing binding regulations, the former is unrepeatable. (shrink)
